The 3700 DNA analyzer increased the pace of large-scale sequencing and genotyping projects since its introduction in 1998 by providing industrial-level throughput with 24-hour unattended operation. Used in the majority of large-scale sequencing laboratories around the world, the 3700 system generates the fundamental genetic map of living organisms.
The acquisition of the 3700 systems underscores deCODE's position as one of the largest and most advanced genotyping facilities in the world. deCODE expects its average genotyping throughput to increase nearly fivefold to approximately 12 million genotypes per month.
Together with its unrivalled genealogical resources and proprietary bioinformatics tools, deCODE uses genotypic analysis of DNA samples from volunteers to identify genes linked to more than three dozen of the world's most common and devastating illnesses. deCODE and its partners use this information to identify new drug targets and to develop more effective treatments.
The ABI PRISM 3700 DNA Analyzer is a fully automated, multi-capillary electrophoresis instrument designed for use in production-scale DNA analysis. It can automatically analyze multiple runs of 96 samples, which enables 24-hour unattended operation.

Because it is a capillary electrophoresis instrument, there is no need to manually pour or load gels. Benchmarking shows length of reads as long as 550 base pairs with 98% accuracy, depending on template and primer quality. The 3700 is an ideal platform for sequencing PCR products less than 550 base pairs in length. It also works well for single stranded and double stranded plasmid DNA templates.
Sample loading, electrophoresis, data collection, and data analysis has been automated to allow unattended operation. The 3700 is a capillary electrophoresis system that can be used to sequence up to 96 DNA templates per run by accommodating up to four 96-well sample plates. A robotic arm loads the samples onto the capillaries; and electrophoresis is completed within five hours. The ABI 3700 can run sequencing reactions based on dye-labeled terminator chemistry and can run three colors of microsatellites, or other fragments, and a size marker in a single lane.
This instrument also utilized uncoated capillaries for higher reliability. Sheath flow technique detects labeled DNA in solution preventing signal loss and background noise caused by light scattering from capillary walls. The ABI 3700 is capable of generating accurate sequence information with as little as 0.5 micrograms of template and can support fragment detection limit of less than 1.0 femtomole. Able to identify 500 bases of a -20M13 primed reaction. Able to detect 200 bases per hour per lane with 98.5% accuracy; detect over 500 bases at 98.5% accuracy at 100 bases/hour. Able to run 96 templates simultaneously with the programmable bypass system. Load and run up to 4 x 96 templates in continuous automated operation with less than 1 hour of instrument attendance/labor per 24 hour period.
The ABI 3700 is compatible with BigDye chemistry which is the state-of-the-art dye-labeled terminator chemistry. Provides data format for analysis as sequence, fragments, assembly into genes and genotypes. These systems are used extensively by the genome centers, Celera, TIGR, etc. to sequence the human genome. Extensive public domain publications allow for easy protocol development.
